VOLUNTARY ANNOUNCEMENT

INTERIM ANALYSIS OF THE PHASE III CLINICAL TRIAL OF PENPULIMAB MONOCLONAL ANTIBODY (PD-1) IN COMBINATION

WITH CHEMOTHERAPY FOR FIRST-LINE SQUAMOUS NON-SMALL CELL LUNG CANCER HAS REACHED KEY ENDPOINTS

This announcement is made by Akeso, Inc. (the "Company", together with its subsidiaries, the "Group") on a voluntary basis to inform the shareholders and potential investors of the Company about the latest business advancement of the Group.

The board of directors of the Company (the "Board") announces that, the interim analysis of the phase III clinical trial of Penpulimab (τ̵̙) (research and development code: AK105), an anti-PD-1 monoclonal antibody drug co-developed by the Company and Sino Biopharmaceutical Limited (stock code: 1177.HK) in combination with paclitaxel and carboplatin for first-line treatment of locally advanced or metastatic squamous non-small cell lung cancer patients, has reached key research endpoints.

Upon the review by the Independent Data Monitoring Committee (IDMC), the interim analysis of the clinical trial demonstrates that, Penpulimab monoclonal antibody in combination with paclitaxel and carboplatin significantly reduces the progressive disease or risk of death as compared with placebo in combination with paclitaxel and carboplatin. Key research endpoints have reached the pre-defined efficacy boundary and the safety profile is consistent with the previously reported study results of Penpulimab monoclonal antibody and no new safety issues were found. Detailed study results will be announced in subsequent academic conferences.

The Company and Sino Biopharmaceutical Limited intend to communicate with the Center for Drug Evaluation (CDE) of the National Medical Products Administration (the "NMPA") of the People's Republic of China (the "PRC") in relation to the drug application for new indications of Penpulimab monoclonal antibody in combination with paclitaxel and carboplatin for first-line treatment of squamous non-small cell lung cancer.

Lung cancer has the highest incidence and mortality among malignant tumors in the PRC and non-small cell lung cancer accounts for approximately 80% of the total lung cancer population.

INFORMATION ABOUT PENPULIMAB (τ̵̙)

Penpulimab (AK105, PD-1 monoclonal antibody) is jointly developed and commercialized by a joint venture established by the Company and Chia Tai-Tianqing Pharmaceutical Group Co., Ltd. ("Chia Tai-Tianqing"), a subsidiary of Sino Biopharmaceutical Limited (stock code: 1177.HK) (together with its subsidiaries "Sino Biopharm"). Penpulimab's fragment crystallisable ("Fc") receptor and complement mediated effector are completely removed by mutations of Fc region, it also has a slower antigen binding offrate compared with the PD-1 antibodies that are already launched in foreign market. These features have made Penpulimab more effective in blocking the activity of the PD-1 pathway and evaded the T-cell anti-tumor activity, thus it has the potential to become an anti-PD-1 drug that can achieve better clinical efficacy.

In May 2020, the NMPA has accepted the new drug application of Penpulimab monoclonal antibody for the treatment of patients with classical Hodgkin's lymphoma (cHL) that has relapsed or refractory (r/r) after two or more lines of systemic chemotherapy (r/r cHL). The registrational clinical trial for third-line metastatic nasopharyngeal cancer with Penpulimab has also met key endpoints.
